Phytochrome Regulation of RNA Synthesis in Isolated Coconut Nuclei

Since the nucleus is the repository of genetic information an investigation was carried out to study the involvement of phytochrome in the control of RNA synthesis in isolated coconut nuclei. Incorporation of 32P-phosphate and 3H-uridine in RNA was reduced by red light (R), and farred light (FR) reversed this effect. RNase activity of the nuclei increased with R but decreased with FR. DNA-RNA hybridization studies showed that DNA template availability for RNA transcription was reduced by R, and FR irradiation nullified the inhibitory effect of R. The RNA molecules synthesized in darkness, R, and FR were different qualitatively as indicated by competitive DNA-RNA hybridization studies.
