Analysis of photosystem II using particle II preparation II. Action spectra for Hill activities and fluorescence properties of variously-treated particle II preparations

  1. To test the scheme proposed in a preceding paper (2), the action spectra for Hill activities and fluorescence properties were measured using variously-treated particle II preparations (P-II).
  2. The action spectrum for ferricyanide photoreduction by intact P-II was characterized by two clear peaks at approximately 686 nm and 648 nm, whereas that for DPIP photoreduction had only one peak at about 686 nm.
  3. The two peaks in the action spectrum for ferricyanide photoreduction by intact P-II were significantly influenced by various treatments; Tris-treatment and antimycin A inhibition resulted in loss of the 686 nm-peak, whereas preillumination treatment and CCCP inhibition brought about the disappearance of the 648 nm-peak.
  4. Aging treatment produced a characteristic effect. The 648 nm-peak diminished gradually with the progress of aging, and only the 686 nm-peak was observed after aging beyond 70 hr.
  5. Fluorescence properties of variously-treated P-II were examined: Aging treatment resulted in the disappearance of the fluorescence of the 730 nm-component.
  6. These findings provide further support for the idea that the two photoreactiorts described above are involved in photosystem II of green plant photosynthesis. Possible roles of the two photoreactions in photosystem II are discussed.
