Membranes carrying acid hydrolases in pea seedling roots
Laboratory of Biochemistry, Faculty of Agriculture, Nagoya University Nagoya, Aichi 464, Japan
Subcellular localization of acid hydrolases in pea seedling roots was studied by differential and sucrose density gradient centrifugations. Significant parts of hydrolase activities in the tissue were recovered in mitochondrial and microsomal fractions. Sedimentable phosphatase was separated into two subtractions: denser and lighter membrane fractions. The distribution of phosphatase activity after sucrose density gradient centrifugation of the denser fraction coincided with that of antimycin Ainsensitive NADH-cytochrome c reductase activity. Electron microscopic observations suggested that the fraction contained only microsomes. RNase in the denser fraction seemed to associate with ribosomes. Phosphatase and RNase were solubilized by sonic treatment in the presence of high concentrations of salt. On the other hand, a-amylase was tightly bound to a membrane. The results are discussed with special regard to the relationship between the membranes and lysosomes.
